Lynk Pharmaceuticals, a clinical-stage innovative drug company headquartered in Hangzhou, China, with operations in Shanghai and Boston, announced on October 27 that its pivotal Phase III trial of zemprocitinib, an investigational selective second-generation oral JAK1 inhibitor, in moderate-to-severe active rheumatoid arthritis produced positive long-term results. The trial enrolled adult patients who had an inadequate response or intolerance to biologic disease-modifying antirheumatic drugs; 430 patients were randomized and 429 received at least one dose of treatment. During the initial 24-week double-blind period, patients received zemprocitinib 12 mg twice daily or placebo. The trial met its primary endpoint of ACR20 response and key secondary endpoints at Week 24, with statistically significant improvement over placebo; results at Week 52 and Week 76 further showed continued improvement in disease activity measures and physical function, with efficacy maintained through Week 76. Rheumatoid arthritis is a chronic autoimmune disease in which persistent joint inflammation can cause pain, functional limitation and disability, and patients who respond inadequately to existing biologics particularly lack effective oral treatment options. The company focuses on innovative drugs for immune and inflammatory diseases, and Boston is one of the key bases of its U.S. operations. The long-term results announced this time show that clinical responses to the drug were maintained through Week 76 of long-term treatment.
